Nic Sestaret scored one of Exeter's five tries on his return as his old team Plymouth Albion went down in front of a record 6,871 crowd at Brickfields.
First-half tries from flanker Tom Johnson and lock Lewis Stevenson helped earn the Chiefs a 20-14 interval lead.
Martin Rice got Albion's one try, aided by three Keiran Hallett penalties.
But second-half tries from wingers Josh Drauniniu and Sestaret, plus centre Matt Cornwell, all converted by Gareth Steenson, completed a comfortable win.
Steenson had also converted both first-half tries, along with two penalties, to end up with a 16-point individual haul.
Exeter's win keeps them in second place, five points behind leaders Leeds, who enjoyed a big away win of their own, 52-7 at Sedgley Park.
Albion drop three places to 10th , thanks to Rotherham Titans' 32-28 win at Cornish Pirates on Sunday.
